A roux is flour cooked into fat before the rest of the dish exists, sometimes for the better part of an hour. A slurry is raw starch whisked into cold water and stirred in during the last minute of cooking, contributing nothing but thickening power. That gap in timing is the whole difference. Everything else — flavor, how long it takes to work, how clear the finished sauce looks — follows from when each one enters the pot and what it's mixed with first.
Fat disperses starch; cold water just makes it wait
Both roux and slurry solve the same problem: raw starch dumped straight into a hot liquid clumps, because the outer layer of each granule gelatinizes on contact and seals before water reaches the center. Fat and cold water solve it by getting between the granules before that can happen — coating them in one case, keeping them apart and cool in the other.
Fat works at any temperature, so a roux can sit on the stove being cooked slowly, its granules held apart by butter or oil, while heat also breaks down some of the flour's structure and browns it — a separate, useful side effect that trades away some thickening power for flavor. Cold water buys no such time. Below roughly 140°F (60°C), cornstarch granules stay inert in suspension, neither swelling nor combining with anything, which is why a slurry can be mixed ahead and left sitting without doing anything at all — until it hits a hot pot, where every granule crosses the gelatinization threshold within seconds of each other and the liquid thickens almost all at once.
The ratio each is built on
A roux starts from equal weights of flour and fat — a tablespoon of butter to a tablespoon of flour is close enough for most kitchens, and running short mid-batch means adding more of both together, never flour alone. A slurry has no fixed ratio in the same way, but a workable starting point is one tablespoon of cornstarch whisked into one tablespoon of cold water, enough to thicken about a cup of hot liquid to a light, glossy gravy. Double both for something heavier.
Why cornstarch thickens harder than flour
Cornstarch is close to pure starch. Flour is only about three-quarters starch by weight — the rest is protein and fiber that do nothing for thickening and scatter light instead of letting it through. That's why a cornstarch slurry needs roughly half the amount, by weight, that a flour-based roux does to thicken the same pot, and why its result comes out clear and glossy rather than opaque. A roux's cloudiness isn't a flaw to fix. It's what makes gravy look like gravy instead of looking like a glaze.

Why a 30-minute gumbo gets a roux and a one-minute glaze gets a slurry
Look at what each one is actually for and the split lines up cleanly. Gumbo's roux gets twenty to thirty minutes over medium-low heat because the dish wants the toasted, almost smoky flavor that a long-cooked roux builds — the thickening is close to a side effect at that point. A béchamel-style sauce for mac and cheese cooks its roux for barely a minute, just long enough to lose the raw-flour taste, because there the goal is body without much flavor of its own.
A teriyaki glaze, a mapo tofu sauce, the gravy under chicken and dumplings — none of them have fifteen spare minutes, and none of them want a flavor from the thickener. Cornstarch mixed with a splash of cold water and stirred in at the end gives them shine and body in under a minute, with the pan sauce or braising liquid doing all the actual flavor work beforehand. Reach for a roux when the thickener has time to become part of the dish. Reach for a slurry when it just needs to finish one.
